Meiringspoort Pass in the Southern Cape, is temporarily closed to traffic.
This follows earlier closures due to flooding and rockfalls.
Motorists are urged to use alternative routes, and proceed with caution.
The Greater Oudtshoorn Municipality said although mop-up operations are underway, the pass is not safe for travel.
Approximately 50 people are currently unable to exit the Meiringspoort Pass.
A reconnaissance helicopter flight is underway to assess the situation.
It is anticipated that vehicles will be able to safely exit Meiringspoort on Saturday morning, once the water level of the Groot River has subsided.
Motorists are urged to use alternative Tradouws Pass, and the via Aberdeen > Willowmore > De Rust route, from Beaufort West.
