An ambulance was among several vehicles stuck in the snow, trying to get to a rescue operation on the N2 near Kokstad earlier on Tuesday.
Authorities from the Kokstad Local Municipality said the N2 to Nolangeni towards Emaxasibeni, is now open, after it was temporarily closed, for safety reasons.
Motorists are urged to drive with caution, on the slippery road or delay trips, until the weather clears.
Roads still closed around Kokstad are the R56 road, between Kokstad and Matatiele, the R617 roads between Kokstad, Swartberg and Underberg and the N2 road from Kokstad via Ingeli.
The N2 near Singisi is also currently closed, due to severe weather conditions and related disruptions.
The South African Weather Service (SAWS) has warned against isolated showers along the east coast of the Eastern Cape, accompanied by damaging winds and waves.
